Self-Nanoemulsifying Systems: An Emerging Technology to Address the Solubility Issues of Drugs

A majority of the new drug candidates display low aqueous solubility, which leads to poor bioavailability, high intrasubject/intersubject variability and lack of dose proportionality. Self-nanoemulsifying drug delivery systems are reported to increase surface area of the drugs to improve solubilization behavior as well as permeation and reduced variability due to effect of food, protection of sensitive drug substances, reduced dosing frequency and better thermodynamically stable state. The aim of this review is to provide an overview of the existing self-nanoemulsifying formulations. The presentation of the various physico-chemical and biopharmaceutical aspects should assist in the understanding of this fascinating field and elucidate the main considerations involved in designing and characterizing a self- nanoemulsifying systems.
