  Recent Progress in the Synthesis of Oxadiazoles

Oxadiazole, a five membered heterocyclic ring has attracted a wide attention to the medicinal chemist in search for the new therapeutic entities. The derivatives of oxadiazole nuclei showed diverse biological activities such as antimicrobial, anti-inflammatory, antitubercular, anticonvulsant, anticancer, anti-HIV and hypoglycemic. Out of its four possible isomers, particularly 1,3,4-oxadiazole derivatives exhibit wide range of applications including pharmacological. A numbers of therapeutic agents containing1,3,4-oxadiazole moiety are available such as nitrofuran antibacterial furamizole, HIV-integrase inhibitor raltagravir, a potent PDF inhibitor BB-83698, antihypertensive agents tiodazosin and nesapidil. The 1,3,4-oxadiazole undergoes number of reactions including electrophillic substitution, nucleophilic substitution, thermal and photochemical. The present review attempts to summarize the various routes of synthesis of 1,3,4-oxadiazole and its derivatives along with their biological potential.
